TBE Buffer 20x Concentrate.
Preparing TBE buffer:
- TBE Buffer Solution for gel electrophoresis of DNA is a 20X concentrate of 100mL volume that should be diluted to produce a working buffer solution.
- To prepare your 1x working solution, redissolve any precipitated salts that may be present by warming the concentrate, then dilute the 20x concentrate by a factor of 20.
For example,
Vol of 20X Concentrate | Volume of Distilled Water | Volume of Working Buffer (1X conc) |
25mL | 475mL | 500mL |
50mL | 950mL | 1,000mL |
100mL | 1,900mL | 2,000mL |
Note: If the TBE has precipitated so much that it has become white throughout, you will need to boil the whole bottle for around fifteen minutes to redissolve.
